FAP Tagging Vectors

We are developing a set of tagging vectors to make it easy to create your own FAP-fusion proteins.  The first available tagging vector is based on Sigma's 3xFLAG vector, and allows for easy creation of an N-Terminal FAP tag.  The vector map is shown below.

This vector codes for a FAP that is a tight binder of the se-Red™ series of fluorogenic dyes, as well as binding the ultra bright Dyedrons™.

By itself, the vectors expresses a cytosolic FAP protein that can activate se-Red-s (the cell permeant fluorogen).  The figure below shows confocal imaging of lipofectamine transfected HEK cells (excitation 640, emission 680lp).  This is a useful control of gene expression prior to creating the fusion utilizing the multi-cloning site.

pFAP-CMV transfected (as-is) into HEK cells.

We bundle 10μg of the plasmid (so you won't need to do a prep right away), along with enough se-Red-s fluorogen to test 5 96 well microtiter plates.  Additional dye can be purchased in 1, 10 and 100 plate vials.  Volume discounts are available.

The N-Terminal pFAP-L5-CMV plasmid + fluorogen is priced at $500.  Contact Us to place an order.

Certain restrictions apply when a Channel Partner is already providing a FAP-based application (GPCR's for example).  If you're looking to create a GPCR-FAP fusion, please contact our partner SpectraGenetics, they have a large selection of GPCRs-FAP fusions ready to go, and will create custom fusions on request.  FLAG is a registered trademark of Sigma corporation.
